红花黄色素对血管性痴呆大鼠学习记忆的影响

摘要

目的 观察红花黄色素对血管性痴呆大鼠学习记忆的影响及相关作用机制。方法 采用双侧颈总动脉结扎致血管性痴呆大鼠模型。水迷宫试验探讨红花黄色素对痴呆大鼠学习记忆的影响, 并用试剂盒测定大鼠皮层及海马组织中超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)和皮层中乙酰转移酶(ChAT)、总胆碱酯酶(T-ChE)的含量。结果 Morris水迷宫实验结果显示, 红花黄色素30 mg·kg-1组、加兰他敏组可明显缩短痴呆大鼠寻找平台潜伏期;空间探索实验中红花黄色素30 mg·kg-1组痴呆大鼠穿越平台次数明显多于模型组;指标测定中, 红花黄色素30 mg·kg-1组可显著升高痴呆大鼠皮层组织中谷胱甘肽过氧化物酶含量和海马组织中谷胱甘肽过氧化物酶、超氧化物歧化酶含量;相较于模型组红花黄色素可以显著降低皮层组织中总胆碱酯酶含量, 升高乙酰转移酶, 但模型组和假手术组比较未见差异。结论 红花黄色素30 mg·kg-1组对血管性痴呆大鼠学习记忆障碍有显著改善作用, 其机制可能与其降低脑组织中氧化应激水平及增加胆碱能神经功能有关。

Abstract

OBJECTIVE The aim of this study is to investigate the effects of safflower yellow(SY)on learning and memory abilitity of vascular dementia (VD) in rats and its possible mechanism of neuro-protective effect against vascular dementia rats. METHODS The bilateral common carotid arteries occluded in two times operation at 3 d interval for vascular dementia model rats. The changes of the spatial learning and memory ability were evaluated by Morris water maze test. The kits of SOD, MDA, GSH-Px, T-ChE, ChAT were used to measure the content of SOD, MDA, GSH-Px, ChAT, T-ChE in cortex and the content of SOD, MDA in hippocampus. RESULTS Learning and memory ability in SY 30 mg穔g-1 group and Galantamine group were better than those in model group. Compared with the model group, GSH-Px level in cerebral cortex, SOD activity and GSH-Px level in hippocampus of VD model rats of SY 30 mg穔g-1 group were significantly enhanced. SY can decrease the level of T-ChE and increase the level of ChAT in cortex, but the differences between model group and sham-operation group were not obvious. CONCLUSION Safflower Yellow could improve learning and memory ability in rats with vascular dementia. Its mechanism might relate to reducing the level of oxidative stress and enhancing the function of cholinergic neurons.
